Conoscenze inerenti la sistematica dei funghi (caratteristiche dei principali taxa fungini). Per gli studenti di Scienze biologiche, tali conoscenze sono fornite attraverso l'insegnamento Diversità vegetale e microbica (terzo anno, SVB0030).

Knowledge concerning the systematics of fungi (characteristics of the main fungal taxa). For students of Biological Sciences, this knowledge is provided through the Plant and Microbial Diversity course (third year, SVB0030).

Obiettivi formativi

L'insegnamento concorre alla realizzazione degli obiettivi formativi dell'ambito ecologico-ambientale del Corso di Laurea in Scienze Biologiche, fornendo conoscenze teoriche e pratiche per l’individuazione ed identificazione dei funghi in ambienti naturali ed artificiali. Attraverso lezioni teorico-pratiche ed esperienze di laboratorio, gli studenti acquisiranno competenze tecniche per la manipolazione dei funghi (isolamento, coltura, trapianti, allestimento di preparati per microscopia ottica) in contesti di ricerca, industriali, biomedici, ambientali. Gli studenti saranno inoltre in grado di interrogare le principali banche dati di sequenze e conosceranno le procedure bioinformatiche per arrivare, a partire da sequenze grezze di DNA, alla descrizione del micobiota desumibile dal sequenziamento high-throughput di marcatori tassonomici. L’insegnamento contribuisce pertanto allo sviluppo della capacità del Biologo junior di eseguire procedure tecnico-analitiche in ambito biotecnologico, biomolecolare, biomedico anche finalizzate ad attivita’ di ricerca.

The teaching contributes to the achievement of the educational objectives of the ecological-environmental area of the Degree Course in Biological Sciences, providing theoretical and practical knowledge for the identification and identification of fungi in natural and artificial environments. Through theoretical-practical lessons and laboratory experiences, students will acquire technical skills for the manipulation of fungi (isolation, culture, subculturing, preparations for optical microscopy) in research, industrial, biomedical and environmental contexts. Students will also be able to interrogate the main sequence databases and will know the bioinformatics procedures to perform, starting from raw DNA sequences, the description of the mycobiota as it may be inferred from the high-throughput sequencing of taxonomic markers. The teaching therefore contributes to the development of the junior biologist's ability to perform technical-analytical procedures in the biotechnological, biomolecular and biomedical fields, also aimed at research activities.

In coerenza con quanto previsto dai Descrittori di Dublino definiti dal Corso di laurea, lo studente dovrà acquisire competenze teoriche e operative in merito a
1) differenze tra approcci coltura-dipendenti ed –indipendenti per la descrizione della diversità fungina nell’ambiente
2) metodi di campionamento per l’analisi del micobiota in matrici ambientali
3) tecniche di manipolazione dei funghi in laboratorio
4) criteri morfologici per il riconoscimento di entità fungine
5) allestimento di librerie di ampliconi destinate al sequenziamento ad alta resa
6) analisi di sequenze singole o multiple di marcatori tassonomici

Consistently with the provisions of the Dublin Descriptors defined by the degree program, the student must acquire theoretical and operational skills regarding
1) differences between culture-dependent and -independent approaches for the description of fungal diversity in the environment
2) sampling methods for the analysis of the mycobiota in environmental matrices
3) fungal manipulation techniques in the laboratory
4) morphological criteria for the recognition of fungal taxa
5) setting up of amplicon libraries for high-throughput sequencing
6) analysis of single or multiple sequences of taxonomic markers

Programma

Differenze tra metodi coltura-dipendenti ed indipendenti per lo studio delle comunità fungine.
Preparazione di terreni colturali (seminaturali e sintetici, generici e selettivi) in uso nel laboratorio micologico; inoculo e propagazione di colture fungine.
Tecniche di conservazione a medio e lungo termine degli isolati fungini. Microcolture su vetrino. Allestimento di preparati microscopici.
Tecniche di isolamento di funghi da suolo (Soil dilution plate).
Identificazione, su basi morfologiche macro- e microscopiche, di rappresentanti dei principali taxa fungini di rilievo come contaminanti di alimenti e manufatti.
Identificazione di isolati fungini mediante barcoding molecolare. Principali marcatori e primers. Estrazione del DNA genomico da colture pure e amplificazione della regione ITS mediante PCR.
Analisi informatica delle sequenze ITS; Utilizzo delle sequenze ITS per l’identificazione delle specie fungine.
Introduzione alle principali tipologie di analisi filogenetica e software di riferimento.
Analisi del micoaerosol in ambienti indoor e outdoor: Principi e tecniche di campionamento. Strategie di campionamento (scelta del campionatore, criteri di monitoraggio).
Aspetti di biodeterioramento fungino dei manufatti (con particolare riguardo ai beni culturali). Tecniche di prevenzione e controllo dello sviluppo fungino (metodi meccanici, fisici e chimici).
Introduzione alle metodiche di valutazione della biodiversità fungina mediante tecniche molecolari di metabarcoding.
Introduzione all’utilizzo delle principali pipeline, software e database per l’analisi del micobiota a partire da sequenze di DNA (MOTHUR, QIIME, USEARCH vsearch, GenBank, UNITE).

Differences between culture-dependent and -independent methods for the study of fungal communities.
Preparation of culture media (semi-natural and synthetic, generic and selective media) for use in the mycological laboratory; inoculation and propagation of fungal cultures.
Medium- and long-term conservation techniques of fungal isolates. Slide micro-cultures. Preparation of slides for microscopic observations.
Soil dilution plate isolation techniques.
Identification, on macro- and micro-scopic morphological bases, of representatives of the main relevant fungal taxa contamining food and manufactured goods.
Identification of fungal isolates by molecular barcoding. Main markers and primers. Genomic DNA extraction from pure cultures and amplification of the ITS region by PCR.

Modalità di insegnamento

L'insegnamento è impartito in lingua inglese.

La frequenza alle lezioni in aula (4 ore) è facoltativa, la frequenza ai laboratori (28 ore) è raccomandata al 100% (eventuali sovrapposizioni con concomitanti insegnamenti, laboratori, esoneri verranno prese in considerazione nella pianificazione del calendario delle attività) e non potrà comunque essere inferiore al 75% come da regolamento del corso di studi.

Qualora perdurassero le restrizioni relative all'emergenza sanitaria Covid-19, l'insegnamento verrà erogato in modalità online. Le lezioni saranno in streaming e le registrazioni, incluse le diapositive utilizzate, saranno caricate sulla pagina Moodle dell'insegnamento.

The teaching is given in English.

Attendance at classroom lessons (4 hours) is optional, attendance at workshops (28 hours) is 100% recommended (any overlap with concomitant courses, workshops, exemptions will be taken into consideration in planning the calendar of activities) and cannot in any case be less than 75% as per the regulations of the course in Biological Sciences.

Should the restrictions relating to the Covid-19 health emergency persist, the teaching will be provided online. The lessons will be streamed and the recordings, including the slides used, will be uploaded to the Moodle page of the teaching.

Modalità di verifica dell'apprendimento

L’esame si svolgerà in forma orale: lo studente verrà valutato mediante 3 domande sulle attività di laboratorio svolte e sulle nozioni impartite durante le lezioni frontali. Il punteggio massimo è 30/30, ogni domanda ha ugual peso (10/30). Prove eseguite in maniera particolarmente efficace vengono premiate con la lode.
Gli studenti con DSA sono invitati a prendere contatto con le docenti per concordare idonee modalità d'esame.

Qualora perdurassero le restrizioni relative all'emergenza sanitaria Covid-19, l’esame verrà condotto in modalità online (WebEx).

The exam will take place orally: the student will be assessed through 3 questions on the laboratory activities carried out and on the concepts taught during the lectures. The maximum score is 30/30, each question has equal weight (10/30). Tests carried out in a particularly effective way are rewarded with honors.
Students with SLD are invited to contact the teacher to agree on suitable examination methods.
Should the restrictions relating to the Covid-19 health emergency persist, the examination will be conducted online (WebEx).
